Abstract
The regulated release of neurotransmitter by exocytosis requires their storage inside specialized, synaptic vesicles. Previous work has ascribed multiple activities and properties to the proteins that transport glutamate into synaptic vesicles, but the relationship between these activities and their physiological role have remained unclear. Intracellular location and low affinity for glutamate have limited the available assays. To circumvent these limitations, we have developed the use of electrophysiology to study the transporters, and use the associated currents to elucidate their mechanism and regulation.
